Redundant and Distinct Roles of Two 14-3-3 Proteins in Fusarium sacchari, Pathogen of Sugarcane Pokkah Boeng Disease

Fusarium sacchari, a key pathogen of sugarcane, is responsible for the Pokkah boeng disease (PBD) in China. The 14-3-3 proteins have been implicated in critical developmental processes, including dimorphic transition, signal transduction, and carbon metabolism in various phytopathogenic fungi. However, their roles are poorly understood in F. sacchari. This study focused on the characterization of two 14-3-3 protein-encoding genes, FsBmh1 and FsBmh2, within F. sacchari. Both genes were found to be expressed during the vegetative growth stage, yet FsBmh1 was repressed at the sporulation stage in vitro. To elucidate the functions of these genes, the deletion mutants ΔFsBmh1 and ΔFsBmh2 were generated. The ΔFsBmh2 exhibited more pronounced phenotypic defects, such as impaired hyphal branching, septation, conidiation, spore germination, and colony growth, compared to the ΔFsBmh1. Notably, both knockout mutants showed a reduction in virulence, with transcriptome analysis revealing changes associated with the observed phenotypes. To further investigate the functional interplay between FsBmh1 and FsBmh2, we constructed and analyzed mutants with combined deletion and silencing (ΔFsBmh/siFsBmh) as well as overexpression (O-FsBmh). The combinations of ΔFsBmh1/siFsBmh2 or ΔFsBmh2/siFsBmh1 displayed more severe phenotypes than those with single allele deletions, suggesting a functional redundancy between the two 14-3-3 proteins. Yeast two-hybrid (Y2H) assays identified 20 proteins with pivotal roles in primary metabolism or diverse biological functions, 12 of which interacted with both FsBmh1 and FsBmh2. Three proteins were specifically associated with FsBmh1, while five interacted exclusively with FsBmh2. In summary, this research provides novel insights into the roles of FsBmh1 and FsBmh2 in F. sacchari and highlights potential targets for PBD management through the modulation of FsBmh functions. The Electric Bus Dynamic Wireless Charging (EB-DWC) system is a bus charging system that enables electric buses to receive power wirelessly from ground-based electromagnetic induction devices. In this system, how to optimally configure the charging infrastructures while considering the unpredictable nature of bus movement is a great challenge. This paper presents an optimization problem for an EB-DWC system in urban settings, addressing stochastic elements inherent in the vehicle speed, initial charging state, and dwell time at bus stops. We formulate a stochastic planning problem for the EB-DWC system by integrating these uncertainties and apply Monte Carlo sampling techniques to effectively solve this problem. The proposed method can improve the system’s robustness effectively. Full article

Isolation, Characterization, and Expression Analysis of NAC Transcription Factor from Andrographis paniculata (Burm. f.) Nees and Their Role in Andrographolide Production

Andrographis paniculata (Burm. f.) Nees is an important medicinal plant known for its bioactive compound andrographolide. NAC transcription factors (NAM, ATAF1/2, and CUC2) play a crucial role in secondary metabolite production, stress responses, and plant development through hormonal signaling. In this study, a putative partial transcript of three NAC family genes (ApNAC83, ApNAC21 22 and ApNAC02) was used to isolate full length genes using RACE. Bioinformatics analyses such as protein structure prediction, cis-acting regulatory elements, and gene ontology analysis were performed. Based on in silico predictions, the diterpenoid profiling of the plant’s leaves (five-week-old) and the real-time PCR-based expression analysis of isolated NAC genes under abscisic acid (ABA) treatment were performed. Additionally, the expression analysis of isolated NAC genes under MeJA treatment and transient expression in Nicotiana tabacum was performed. Full-length sequences of three members of the NAC transcription factor family, ApNAC83 (1102 bp), ApNAC21 22 (996 bp), and ApNAC02 (1011 bp), were isolated and subjected to the promoter and gene ontology analysis, which indicated their role in transcriptional regulation, DNA binding, ABA-activated signaling, and stress management. It was observed that ABA treatment leads to a higher accumulation of andrographolide and 14-deoxyandrographolide content, along with the upregulation of ApNAC02 (9.6-fold) and the downregulation of ApNAC83 and ApNAC21 22 in the leaves. With methyl jasmonate treatment, ApNAC21 22 expression decreased, while ApNAC02 increased (1.9-fold), with no significant change being observed in ApNAC83. The transient expression of the isolated NAC genes in a heterologous system (Nicotiana benthamiana) demonstrated their functional transcriptional activity, leading to the upregulation of the NtHMGR gene, which is related to the terpene pathway in tobacco. The expression analysis and heterologous expression of ApNAC21 22 and ApNAC02 indicated their role in andrographolide biosynthesis. Full article

Adherence to the CLOSE Protocol and Low Baseline Generator Impedance Are Independent Predictors of Durable Pulmonary Vein Isolation

Background: Atrial fibrillation (AF) recurrence after pulmonary vein isolation (PVI) is predominantly attributed to pulmonary vein reconnection (PVR). Predictors of AF recurrence have been widely studied; however, data are scarce on procedural parameters that predict chronic PVR. We aimed to study PVR rates and predictors of PVR. Methods: We retrospectively included 100 patients who underwent repeated ablation due to AF recurrence after initial PVI with the CARTO system. PVR was determined during the repeated procedure by electrophysiological evaluation, and initial procedural characteristics predicting PVR were studied, including adherence to the CLOSE protocol, use of high power, first-pass isolation (FPI), and baseline generator impedance (BGI). Results: Thirty-eight patients underwent initial CLOSE-guided PVI, and sixty-two underwent initial non-CLOSE PVI. A repeat procedure was performed 23 ± 16 months after the initial procedure. In total, PVR was found in 192 of 373 PVs (51.5%), and all PVs were isolated in 17/100 (17%) patients. Factors associated with all PVs being isolated were adherence to the CLOSE protocol, a higher power setting, the presence of bilateral FPI, and lower BGI (88% vs. 28%, p < 0.0001; 37.5 W vs. 30 W, p = 0.0276; 88.2% vs. 40.4%, p = 0.0007; and 127.6 Ω vs. 136.6 Ω, p = 0.0027, respectively). In initial procedures with adherence to the CLOSE protocol, the FPI rate was significantly higher (73.7% vs. 25%, p < 0.0001), while there were no significant differences in terms of procedure time and left atrial dwell time (81 vs. 85 min, p = 0.83; and 60 vs. 58 min, p = 0.08, respectively). BGI ≥ 130 Ω (AUC = 0.7403, sensitivity: 77.1%, specificity: 68.8%, p = 0.0032) was associated with a significantly higher probability of PVR (OR = 6.757; p < 0.0001). In multivariable analysis, independent predictors for PVR were non-adherence to the CLOSE protocol and BGI ≥ 130 Ω. Conclusions: Our findings indicate that adherence to the CLOSE protocol and baseline generator impedance < 130 Ω during AF ablation are independent predictors of PVI durability. Full article

An organism’s ability to function properly depends not solely on its diet but also on the intake of nutrients and non-nutritive bioactive compounds that exert immunomodulatory effects. This principle applies both to healthy individuals and, in particular, to those with concomitant chronic conditions, such as type 2 diabetes. However, the current food industry and the widespread use of highly processed foods often lead to nutritional deficiencies. Numerous studies have confirmed the occurrence of immune system dysfunction in patients with type 2 diabetes. This article elucidates the impact of specific nutrients on the immune system function, which maintains homeostasis of the organism, with a particular emphasis on type 2 diabetes. The role of macronutrients, micronutrients, vitamins, and selected substances, such as omega-3 fatty acids, coenzyme Q10, and alpha-lipoic acid, was taken into consideration, which outlined the minimum range of tests that ought to be performed on patients in order to either directly or indirectly determine the severity of malnutrition in this group of patients. Exploring Geometrical, Electronic and Spectroscopic Properties of 2-Nitroimidazole-Based Radiopharmaceuticals via Computational Chemistry Methods

Tumor hypoxia plays an important role in the clinical management and treatment planning of various cancers. The use of 2-nitroimidazole-based radiopharmaceuticals has been the most successful for positron emission tomography (PET) and single photon emission computed tomography (SPECT) imaging probes, offering noninvasive means to assess tumor hypoxia. In this study we performed detailed computational investigations of the most used compounds for PET imaging, focusing on those derived from 2-nitroimidazole: fluoromisonidazole (FMISO), fluoroazomycin arabinoside (FAZA), fluoroetanidazole (FETA), fluoroerythronitroimidazole (FETNIM) and 2-(2-nitroimidazol-1-yl)-N-(2,2,3,3,3-pentafluoropropyl)acetamide (EF5). Conformational analysis, structural parameters, vibrational IR and Raman properties (within both harmonic and anharmonic approximations), as well as the NMR shielding tensors and spin-spin coupling constants were obtained by density functional theory (DFT) calculations and then correlated with experimental findings, where available. Furthermore, time-dependent DFT computations reveal insight into the excited states of the compounds. Our results predict a significant change in the conformational landscape of most of the investigated compounds when transitioning from the gas phase to aqueous solution. According to computational data, the 2-nitroimidazole moiety determines to a large extent the spectroscopic properties of its derivatives. Due to the limited structural information available in the current literature for the investigated compounds, the findings presented herein deepen the current understanding of the electronic structures of these five radiopharmaceuticals. Lyophilized plant-origin extracts are rich in highly potent antioxidant polyphenols. In order to incorporate them into food products, it is necessary to protect these phytochemicals from atmospheric factors such as heat, light, moisture, or pH, and to enhance their bioavailability due to their low solubility. To address these challenges, recent studies have focused on the development of encapsulation techniques for antioxidant compounds within polymeric structures. In this study, lyophilized olive leaf extracts were microencapsulated with the aim of overcoming the aforementioned challenges. The method used for the preparation of the studied microparticles involves external ionic gelation carried out within a water–oil (W/O) emulsion at room temperature. HPLC analysis demonstrates a high content of polyphenols, with 90% of the bioactive compounds encapsulated. Meanwhile, quantification by inductively coupled plasma optical emission spectroscopy (ICP-OES) reveals that the dried leaves, lyophilized extract, and microencapsulated form contain satisfactory levels of macro- and micro-minerals (calcium, potassium, sodium). The microencapsulation technique could be a novel strategy to harness the polyphenols and minerals of olive leaves, thus enriching food products and leveraging the antioxidant properties of the polyphenolic compounds found in the lyophilized extract. This study explores the use of Halloysite NanoTubes (HNTs) as photocatalysts capable of decomposing organic dyes under exposure to visible or ultraviolet light. Through a systematic series of photocatalytic experiments, we unveil that the photodegradation of Rhodamine B, used as a model cationic dye, is significantly accelerated in the presence of HNTs. We observe that the extent of RhB photocatalytic degradation in 100 min in the presence of the HNTs is ~four times higher compared to that of bare RhB. Moreover, under optimized conditions, the as-extracted photodegradation rate of RhB (~0.0022 min−1) is comparable to that of the previously reported work on the photodegradation of RhB in the presence of tubular nanostructures. A parallel effect is observed for anionic Coumarin photodegradation, albeit less efficiently. Our analysis attributes this discrepancy to the distinct charge states of the two dyes, influencing their attachment sites on HNTs. Cationic Rhodamine B molecules preferentially attach to the outer surface of HNTs, while anionic Coumarin molecules tend to attach to the inner surface. By leveraging the unique properties of HNTs, a family of naturally occurring nanotube structures, this research offers valuable insights for optimizing photocatalytic systems in the pursuit of effective and eco-friendly solutions for environmental remediation. Spatial truss structures (STSs), serving as the bottom support structure of a cooling tower, effectively harness the superior load-bearing capacity offered by lattice-type truss structures. STSs are composed of main bars, diagonal bars, and horizontal bars, with horizontal bars serving as vital components of the truss structure. They play a pivotal role in maintaining the overall integrity and stability of the structure. The proportional relationship between the stiffness of each bar in STSs has a profound impact on the mechanical characteristics of the overall structure. This relationship directly influences the ultimate load-bearing capacity of the structure. Therefore, conducting research on the influence patterns of this relationship is of utmost importance. This paper explores the study of triangular truss structures (TTSs) and quadrilateral truss structures (QTSs). Firstly, through theoretical analysis, considering structural elements such as the stiffness of the horizontal bars, the number of layers in the truss, and the angle between the diagonal bars and the horizontal bars, theoretical expressions for the calculation of the ultimate load capacity of TTSs and QTSs are derived. Furthermore, a parametric finite element (FE) model was established for the TTSs and QTSs. Through numerical simulations, the validity of the theoretical calculation expressions was verified. Finally, this paper discusses the influence of factors such as the stiffness of the horizontal bars, the number of layers in the truss, and the angle between the diagonal and horizontal bars on the TTSs and QTSs. It analyzes the patterns and trends of these influences. The research results indicate that the theoretical and numerical simulation results for the TTSs have an error ranging from 0.40% to 4.93%, while the relative error for the QTSs ranges from 1.59% to 4.88%. These errors are within an acceptable range for engineering calculations. As the stiffness of the horizontal bars increases, the proportionality coefficient of the truss’s ultimate load capacity shows an initial increase followed by a stable trend. It reaches an equilibrium state when the stiffness of the horizontal bars reaches a certain threshold. As the number of layers in the truss and the angle between the diagonal and horizontal bars increase, the proportionality coefficient of the load capacity gradually decreases. The research findings provide a theoretical basis for the application of TTSs and QTSs in cooling towers. Full article

Peripheral and autonomic neuropathy are common disease manifestations in systemic amyloidosis. The neurofilament light chain (NfL), a neuron-specific biomarker, is released into the blood and cerebrospinal fluid after neuronal damage. There is a need for an early and sensitive blood biomarker for polyneuropathy, and this systematic review provides an overview on the value of NfL in the early detection of neuropathy, central nervous system involvement, the monitoring of neuropathy progression, and treatment effects in systemic amyloidosis. A literature search in PubMed, Embase, and Web of Science was performed on 14 February 2024 for studies investigating NfL levels in patients with systemic amyloidosis and transthyretin gene-variant (TTRv) carriers. Only studies containing original data were included. Included were thirteen full-text articles and five abstracts describing 1604 participants: 298 controls and 1306 TTRv carriers or patients with or without polyneuropathy. Patients with polyneuropathy demonstrated higher NfL levels compared to healthy controls and asymptomatic carriers. Disease onset was marked by rising NfL levels. Following the initiation of transthyretin gene-silencer treatment, NfL levels decreased and remained stable over an extended period. NfL is not an outcome biomarker, but an early and sensitive disease-process biomarker for neuropathy in systemic amyloidosis. Therefore, NfL has the potential to be used for the early detection of neuropathy, monitoring treatment effects, and monitoring disease progression in patients with systemic amyloidosis. Full article

Experiential learning methods are advantageous for students as they motivate them to comprehend structural concepts without complex calculations, enhancing their inherent understanding of static principles. This research introduces a novel, cost-effective haptic didactic tool to enhance the approach to teaching trusses to students in a School of Architecture. The primary goal is to address challenges associated with the complexities of teaching structural systems within the context of architectural education. The proposed approach is related to the most critical issue, which is the state in which the individual elements are under applied load, compression, or tension. The approach explores the deformation of the truss elements and establishes a connection between their visible deformation and the stress they develop under various loads. As a didactic tool, this approach offers an alternative perspective to help students understand truss function under various loads. Also, an assessment procedure of learning outcomes and satisfaction indices has been structured to validate the impact on students on the proposed educational procedure. The findings underscore the significant educational efficiency of the proposed procedure as a sustainable way to connect the structural engineering challenges arising during design courses and creative skills in architecture engineering. Background: Some bandages with Kinesiotape have demonstrated some potential as means of prevention for Anterior Cruciate Ligament (ACL) injury in men. Objective: The main objective of this pilot study was to observe if a Kinesiotape bandage at the knee could potentially have an immediate preventive effect against ACL injuries by improving proprioception and balance and reducing knee valgus and anterior translation of the tibia during certain functional tests in female athletes. Materials and Methods: A cross-over clinical trial including 10 female athletes (football and rugby) was conducted, where the two lower limbs of the 10 participants (n = 20) were randomly assigned to the intervention group (IG) or to the control group (CG). A Kinesiotape bandage was placed on one knee with a tension of 75% (IG, n = 10) and another bandage on the other knee with a tension of 10% (CG, n = 10), as a placebo. Ultrasound assessment and functional tests were performed before (T1) and after (T2) bandaging according to group assignment. A videographic analysis was carried out with the ImageJ Software version 2.0. Results: A statistically significant reduction in knee valgus was observed in the Lateral Step Down test in the IG, compared to the CG (p < 0.05; η2p = 0.26) (IG-T1: 151.40 ± 11.04°; IG-T2: 157.10 ± 10.18°; versus CG-T1: 156.96 ± 5.44°; CG-T2: 158.68 ± 6.12°). In the other tests, no significant differences were found in terms of time × group interaction. Conclusions: A Kinesiotape bandage was able to reduce knee valgus in a functional test compared to a placebo bandage but was not more effective at reducing the anterior translation of the tibia or improving balance or proprioception. Based on these results, it could have a modest preventive effect against an ACL injury in female athletes. Full article

Silibinin’s Effects against Methotrexate-Induced Hepatotoxicity in Adjuvant-Induced Arthritis Rat Model

Methotrexate (MTX) is the first drug of choice to treat several diseases, including rheumatoid arthritis. However, its administration is accompanied by severe side effects, most commonly hepatotoxicity. Hence, alternative therapies with a lower toxicity and fewer side effects are needed. This study aimed to investigate the antioxidant and hepatoprotective effects of silibinin (SIL, natural agent) against MTX-induced hepatotoxicity in an adjuvant-induced arthritis (AIA) rat model. Arthritic rats were treated with SIL (100 mg/kg) and/or methotrexate (2 mg/kg). Non-arthritic rats, arthritic untreated rats, and arthritic rats who received the vehicle were followed in parallel. SIL alleviated the systemic consequences of arthritis by restoring lost weight, decreasing the erythrocyte sedimentation rate, and ameliorating joint damage, which was evident both micro- and macroscopically. Additionally, SIL prevented the histopathological alterations in the liver and significantly reduced the liver damage caused by MTX and AIA, as shown by a decrease in the markers of liver damage (ALT and AST). Furthermore, SIL relieved the oxidative stress induced by AIA and MTX in liver tissue by decreasing the lipid peroxidation (MDA) levels and enhancing the antioxidant defense system (GSH levels; catalase and superoxide dismutase (SOD) activities). In conclusion, our results suggest that SIL is a potent antioxidant and hepatoprotective agent in arthritic rats. It markedly attenuated the progression and severity of the arthritic disease and eased the oxidative stress in liver tissue by improving the pro-oxidant/antioxidant balance. Full article

Background: Sodium fluctuation is independently associated with clinical deterioration. We developed and validated a prognostic index based on sodium fluctuation for risk stratification and in-hospital monitoring. Methods: This study included 33,323 adult patients hospitalized at a tertiary care hospital in 2014. The first 28,279 hospitalizations were analyzed to develop the model and then the validity of the model was tested using data from 5044 subsequent hospitalizations. We predict in-hospital mortality using age, comorbidity, range of sodium fluctuation, and duration of sodium fluctuation, abbreviated as CARDS. Results: In-hospital mortality was similar in the derivation (0.6%) and validation (0.4%) cohorts. In the derivation cohort, four independent risk factors for mortality were identified using logistic regression: age (66–75, 2 points; >75, 3 points); Charlson comorbidity index (>2, 5 points); range of sodium fluctuation (7–10, 4 points; >10, 10 points); and duration of fluctuation (≤3, 3 points). The AUC was 0.907 (95% CI: 0.885–0.928) in the derivation cohort and 0.932 (95% CI: 0.895–0.970) in the validation cohort. In the derivation cohort, in-hospital mortality was 0.106% in the low-risk group (0–7 points), 1.076% in the intermediate-risk group (8–14 points), and 8.463% in the high-risk group (15–21 points). In the validation cohort, in-hospital mortality was 0.049% in the low-risk group, 1.064% in the intermediate-risk group, and 8.403% in the high-risk group. Conclusions: These results suggest that patients at low, intermediate, and high risk for in-hospital mortality may be identified by CARDS mainly based on sodium fluctuation. Full article

The aim of this work is to analyze the effect of water absorption on the mechanical properties and damage mechanisms of polyester/glass fiber/jute fiber hybrid composites obtained using the compression molding and vacuum-assisted resin transfer molding (VARTM) techniques with different stacking sequences. For this purpose, the mechanical behavior under tensile stress of the samples was evaluated before and after hygrothermal aging at different temperatures: TA, 50 °C, and 70 °C for a period of 696 h. The damage mechanism after the mechanical tests was evaluated using SEM analysis. The results showed a tendency for the mechanical properties of the composites to decrease with exposure to an aqueous ambient, regardless of the molding technique used to conform the composites. It was also observed that the stacking sequence had no significant influence on the dry composites. However, exposure to the aqueous ambient led to a reduction in mechanical properties, both for the molding technique and the stacking sequence. Damage such as delamination, fiber pull-out, fiber/matrix detachment, voids, and matrix removal were observed in the composites in the SEM analyses. Full article

The application of computer vision and machine learning methods for semantic segmentation of the structural elements of 3D-printed products in the field of additive manufacturing (AM) can improve real-time failure analysis systems and potentially reduce the number of defects by providing additional tools for in situ corrections. This work demonstrates the possibilities of using physics-based rendering for labeled image dataset generation, as well as image-to-image style transfer capabilities to improve the accuracy of real image segmentation for AM systems. Multi-class semantic segmentation experiments were carried out based on the U-Net model and the cycle generative adversarial network. The test results demonstrated the capacity of this method to detect such structural elements of 3D-printed parts as a top (last printed) layer, infill, shell, and support. A basis for further segmentation system enhancement by utilizing image-to-image style transfer and domain adaptation technologies was also considered. The results indicate that using style transfer as a precursor to domain adaptation can improve real 3D printing image segmentation in situations where a model trained on synthetic data is the only tool available. The mean intersection over union (mIoU) scores for synthetic test datasets included 94.90% for the entire 3D-printed part, 73.33% for the top layer, 78.93% for the infill, 55.31% for the shell, and 69.45% for supports. The assessment of iron levels in cattle muscle tissue is crucial for livestock management because it influences both animal health and meat quality, key factors in sustainable development. This study aimed to develop an optimal model for noninvasively predicting the iron content in Hereford cattle muscle tissue, contributing to a comprehensive understanding of the animals’ elemental status. The research involved the atomic absorption analysis of muscle tissue and hair samples from cattle. A regression model was constructed using the least squares method to identify the most effective approach. These findings have ecological applications, aiding in evaluating environmental health and establishing acceptable iron thresholds for animals. The proposed mathematical model utilizing biomarkers (levels of Mg, K, Fe, Al, Cr in hair) will allow for the assessment of iron levels in cattle muscle tissue throughout the period of productive use, with the possibility of adjustment and tracking the changes in elemental status over time. The utilization of the developed method will enable the diagnosis of animal elementosis and assessment of the iron level burden. Subsequently, this will allow for the improvement of the qualitative characteristics of the final product. Thus, the obtained data contribute to fundamental knowledge regarding the content and variability of iron levels in the muscle tissue of cattle. Full article

This study investigates the interplay between emotional competence, self-efficacy, and teaching experience in determining preschool teachers’ performance. Drawing on Bandura’s Theory of Self-Efficacy, Goleman’s Emotional Intelligence Theory, and Bronfenbrenner’s Ecological Systems Theory, the research employs a quantitative approach, analyzing responses from preschool teachers in Zhejiang province, China. Key findings reveal that emotional competence significantly predicts teachers’ performance and self-efficacy, with self-efficacy further mediating this relationship. Interestingly, while teaching experience moderates the impact of emotional competence on self-efficacy, it does not significantly influence the relationship between self-efficacy and teaching performance. The study underscores the critical role of emotional competence in teaching efficacy and highlights the complexity of how teaching experience interacts with these dynamics. These insights are crucial for developing targeted interventions in teacher training programs, emphasizing emotional skills and self-belief as key drivers of effective teaching in early-childhood education. Currently, fiber-reinforced polymer composites (FRPs) used for demanding structural applications predominantly utilize carbon, glass, and aramid fibers embedded in epoxy resin, albeit occasionally polyester and vinyl ester resins are also used. This study investigates the feasibility of employing recyclable and sustainable materials to formulate a composite suitable for load-bearing structural applications, particularly in scenarios involving low-velocity and high-velocity impacts (LVIs and HVIs, respectively). The paper presents a comparative analysis of the performance of basalt–Elium, a fully recyclable, sustainable, and environmentally friendly composite, with an epoxy-based counterpart. Moreover, an accurate and reliable numerical model has been developed and introduced through which the response of these composites can be examined efficiently and accurately under various loading states. The results of this investigation demonstrate the viability of the basalt–elium composite as a fully recyclable and sustainable material for crafting efficient and lightweight composites. Additionally, the accurately developed finite element model presented here can be used to assess the influence of several parameters on the composite, thereby optimizing it for a given situation. The gut microbiota plays a significant role in tumor pathogenesis by regulating the host metabolism and immune response, and there are few studies focused on tracking changes in the gut microbiota from the onset of lung cancer. Therefore, the aim of our study is combining preclinical and clinical research to thoroughly analyze the signatures of fecal microbiota in lung cancer, which will be useful for early diagnosis and predicting the therapeutic efficacy of lung cancer. The first part of this study analyzed the fecal metagenomic differences between patients with non-small cell lung cancer and healthy subjects, and the second part of this work constructed a murine lung cancer model to monitor changes in mouse fecal metagenomics and T cell immunology during lung cancer progression. We found that the fecal microbiota was altered in both humans and mice with lung cancer, characterized by a significantly reduced microbial diversity and number of beneficial microbes, with increases in potential pathogens. The fecal level of Akkermansia muciniphila and the gut metabolic module of the secondary bile acid metabolism were diminished in both humans and mice with lung cancer compared with healthy subjects. Splenomegaly was observed in the lung cancer mice. Flow cytometer analysis of the splenocytes revealed substantial alterations in the proportions of T cell subsets in the lung cancer mice, characterized by significant increases in CD4+Foxp3+CD25+ T regulatory cells (p < 0.05) while significant decreases in CD3+ T cells (p < 0.001), CD4+ T cells (p < 0.001), and the CD4+/CD8+ ratio (p < 0.01). Vertical and longitudinal analyses of the fecal microbiota of the two mouse groups identified some lung cancer biomarkers (including Acutalibacter timonensis, Lachnospiraceae bacterium NSJ-38 sp014337195, etc.). The fecal microbiota of the lung cancer mice had a reduced metagenomic potential for neurotransmitters (melatonin, γ-aminobutyric acid, and histamine) compared with healthy mice. In summary, this study found that the diversity, structure, and composition of gut microbiota vary between cancer and healthy conditions, ultimately leading to changes in the potential for functional metagenomics. Background: Androgenetic alopecia, the most common cause of non-scarring hair loss, is a consequence of the gradual miniaturization of the hair follicles. In the majority of male androgenetic alopecia cases, a patient’s history and clinical evaluation may be sufficient to establish the diagnosis, while for women, they should be supplemented with trichoscopy. Methods: The PubMed and Scopus databases were used to collate published studies and to analyze the most typical trichoscopic findings in patients diagnosed with androgenetic alopecia. A total of 34 articles were retrieved after exclusion. Results: The most common features identified using trichoscopy included hair diameter variability (94.07% of patients), vellus hairs (66.45%) and the peripilar sign (43.27%). Others, such as the honeycomb pattern, yellow and white dots, were less relevant. Conclusions: We concluded that hair diameter variability, vellus hairs and the peripilar sign represented valuable indicators for the diagnosis of androgenetic alopecia. Full article

Experimental and Simulation Investigation on Fatigue Performance of H13 Steel Tools in Friction Stir Welding of Aluminum Alloys

As the central component in friction stir welding, the design and manufacture of welding tools for aluminum alloys have garnered substantial attention. However, the understanding of tool reliability during the welding process, especially in terms of fatigue performance, remains unclear. This paper focuses on the welding of AA2219-T4 as a case study to elucidate the predominant failure mode of the tool during the friction stir welding (FSW) of aluminum alloys. Experimental methods, including FSW welding and fracture morphology analysis of the failed tool, coupled with numerical simulation, confirm that high-cycle mechanical fatigue fracture is the primary mode of the tool failure. Failures predominantly occur at the tool pin’s root and the shoulder end face with scroll concave grooves. The experimental and simulation results exhibit a noteworthy agreement, validating the reliability of the simulation model. The FSW Arbitrary Lagrangian–Eulerian (ALE) model developed in this study analyzes stress distribution and variation under the thermo-mechanical coupling effect of the tool. It reveals that stress concentration resulting from structural changes in the tool is the primary driver of fatigue crack initiation. This is attributed to exposure to alternating cyclic stresses such as bending, tension, and torsion at the tool pin’s root, manifesting as multiaxial composite mechanical fatigue. Among these stresses, bending alternating cyclic stress exerts the most significant influence. The paper employs the Tool Life module in DEFORM software to predict the fatigue life of the tool. Results indicate that reducing welding speed or increasing rotation speed can enhance the tool’s fatigue life to some extent. The methodology proposed in this paper serves as a valuable reference for optimizing FSW structures or processes to enhance the fatigue performance of welding tools. Full article

Soil nailing is a prevalent and cost-effective technique employed to reinforce and enhance the stability of precarious natural or cut slopes; however, its application as a primary support system to prevent collapses or cave-ins during foundation excavation could be more frequent. To better understand the behavior of such a support system, this study simulated a full-scale nail-supported excavation for the foundation pit of a 20-story building to examine the effect of placing a strip footing with various combinations of configurations on the crest of the excavation pit. The results are discussed in terms of the nail axial force, wall horizontal deflection, basal heave, and safety factor against sliding. The results show that the footing width and setback distance are the two most significant factors dominating the wall horizontal deflection. This study also reveals that the maximum axial force is closely related to the apparent active earth pressure, which accounts for the presence of a tension crack, at nail depth. Such a finding allows engineers to assess and mitigate the risks of structural failure more effectively and optimize the design of nail-retaining structures. Full article
